General Information

Two biomaterial portions from a single urination are examined to detect organized and unorganized sediment and cylindruria.

Indications

Biomaterial is collected to determine pyuria’s presence and quantity for localization and to identify other urological conditions (inflammation, infections) via microscopic examination, turbidity analysis, and leukocyte counts in the second portion.

Since urine contains kidney metabolic products, it’s ideal for diagnosing urethral and adrenal issues. The two-glass test requires adherence to daily diuresis, collected in the morning when urine concentrates overnight, providing specialists with an objective picture.

Preparation for the Test

Genital hygiene is crucial for the two-glass test. Avoid soap or chemical products. Prepare a sterile container (up to 500 mL), available at pharmacies.

How the Test Is Conducted

Urination occurs in two stages (two containers). The first holds up to 50 mL; the second, the remainder. If unwanted samples are found only in the 50 mL container, it indicates urethral inflammation. If in the second, it suggests prostate or seminal vesicle involvement (in men). Gnostic presence in the two-glass test indicates pathology and damage.

Two-glass test norms should be:

• White blood cells – women: up to 6, men: up to 3; children – girls: up to 6, boys: up to 4;
• Clotted protein – singular.

Interpretation

Elevated bladder epithelial cells suggest cystitis or nephropathy; renal pelvis cells – inflammation or urethritis; blood – micro/macrohematuria or erythrocyte leakage in the urinary system. Elevated leukocytes indicate leukocyturia (up to 20) or pyuria (up to 60). Excess hyaline cylinders signal fever, renal issues, or proteinuria.

Special Considerations

The two-glass test is indicated monthly for pregnant women at various fetal development stages. Values align with norms, but late-detected issues may harm the fetus, requiring careful preparation.
