General information

To the main diagnostic methods include:

  • microscopic;
  • cultural;
  • luminescent;
  • histological;
  • immunological.

Procedure

In therapeutic practice, the diagnosis of mycoses at the initial stages is limited to microscopic and cultural studies from the point of view of necessity and expediency.

Microscopic

It is performed with a standard set of microscopy tools (microscopes), for which the structures obtained by cutting or scraping are revealed as a native or stained substance with subsequent clarification (maceration).

It reveals budding cells and filiform hyphae and allows judge only the nature of the infection, without giving a complete picture of the source of the contagion itself. The performance indicator is determined by a significant share of error and directly depends on the level and qualifications of the personnel.

Cultural

A fairly sensitive method of detection and recognition, allowing you to identify and determine the type of contagion even with negative microscopy results. It is considered appropriate for recognizing latent forms of mycoses and dermatophytes transmitted by healthy people.

The sowing is performed according to the algorithm - first in Sabouraud's medium, in the presence of antibiotics that suppress the growth of contaminants entering with the air, and then reseeding the grown cultures in selective media.

The updating process is completed by microscopic analysis.

The purity of positive results varies within 30-50%, subject to compliance with the requirements of the norms and rules for sowing, collecting and processing materials, on high-quality equipment by highly qualified specialists.

Luminescent

This method is based on a physical phenomenon - the glow effect of hair affected by spores of the Microsporum fungus, observed when passing rays in the ultraviolet range of wavelengths (365 nm) through a Wood fluorescent filter with the material under study.

Unfortunately, this phenomenon is effective only in material completely affected by the fungus. In the early stages of fresh lesions, the effect may be absent or partially offset by side factors. Subsequently, the pathology of the identified lesions is additionally subjected to microscopy.

Immunological

The purpose of such technology is authentic identification of the specificity of the body's reaction, taking into account the serological component of the fungal infection.

Antibodies are detected by appropriate reactions of serum samples with antigens.

Cellular and humoral immunity is assessed by the results of tests (Pirquet, Moro or Mantoux) for allergic reactions.

The final comparison of diagnostics provides an up-to-date reliable perception and predictable dynamics.

The biological component is a special complex of laboratory studies of the most dangerous and deep pathologies of infection, to which experimental animals are subjected, with constant monitoring of the course and development of the disease.

Histological

The introduction of fungi into epidermis and stratum corneum is non-specific for the organism and causes a response inflammatory immune reaction with pathomorphological changes in the foci.

The technique of diagnostics with histological preparations (changing color) is capable of detecting polysaccharides of chitin and cellulose in the cell walls of most known dermatophyte fungi. Among the most common and progressive are PAS (periodic acid exposure).

Indications

The material for research is hair, scales, vesicle covers, nail plates, epidermal particles that have the most frequent and erased manifestations of epidermophytosis and rubrophytosis.

The basis for laboratory diagnostics of mycoses is a fairly clear visualization of the signs of clinical manifestations and the appointment of a course by a specialized specialist.

No side effects are observed when performing the procedures.
