General Information

Connective tissue (skin, muscles, joints) consists of protein fibers rich in collagen, providing softness and flexibility. In systemic diseases affecting these tissues, the body produces autoantibodies against its own proteins. The immune system attacks its own cells, leading to their deformation, destruction, and impaired organ function.

In SLE, skin is primarily affected, with facial rashes, hair loss, anemia, and impaired memory/concentration.

The main autoantibodies in SLE target dsDNA. Immune complexes formed during their interaction with deoxyribonucleic acid accumulate on vessel walls, organs, and skin cell membranes, triggering inflammatory and toxic reactions.

Differential diagnosis considers not only their presence but also titer and production persistence.

The test determines disease activity: high levels indicate progression, low levels suggest remission. Anti-DNA levels are monitored every 3-6 months in mild cases.

A negative result excludes SLE in 98% of cases and in 87% of patients with other autoimmune conditions.

Elevated anti-dsDNA levels correlate with circulating immunoglobulin (IgG) complexes in renal glomeruli due to the close interaction of double-stranded polymer chains with renal cell membranes.

Indications for Anti-dsDNA (Native) Antibody Testing

The test is indicated for suspected SLE with symptoms such as:

  • Rashes on cheeks, nose bridge, chest, and forearms;
  • Persistent subfebrile temperature (37-37.5°C), chills, and fever;
  • Joint stiffness, lung inflammation, pericarditis;
  • Renal dysfunction.

Other indications include:

  • Presence of antinuclear antibodies in serum;
  • Monitoring treatment efficacy.

How Anti-dsDNA (Native) Antibody Testing Is Conducted

No special preparation is required. Avoid smoking 30 minutes prior. A specialist draws venous blood using the enzyme-linked immunosorbent assay (ELISA) method.

Elevated antibody levels without SLE symptoms suggest possible disease development, warranting rheumatologist follow-up and repeat testing.
